Confocal Laser Scanninng Microscope, Zeiss LSM 510

 

 

The laser module has 3 single photon lasers (Ar, HeNe 543, HeNe 633) which allow excitation lines of 458, 488, 514, 543, 568, and 633 nm. The detector module has 3 confocal detectors (photomultiplier tubes with pinholes). The scan module can be fitted on either the Axiovert 200M or the Axioplan 2, both fully motorised microsopes controlled via a standard Pentium PC. The Axioplan motorised research microscope is equipped with 3 Plan Neofluar objectives; 10X, 20X, 50X The Axiovert 100M SP motorised research microscope is equipped with 3 Plan Neofluar objectives; 10X, 20 X, 40X (1.3NA oil) and a Plan Apochromatic objective; 63X (1.4 NA oil) All objective lenses are ICS lenses (infinity colour corrected system) Both microscopes are configured for transmitted light, brightfield, phase contrast, fluorescence (filter sets for UV, blue & green excitation), reflected light, DIC (differential interference contrast).

The Zeiss LSM software, which runs on Windows NT 4.0 Operating System, provides a wide range of image processing functions,
including all standard 2D/3D functions (stereo,projection, depth colour coding,viewing from any angle), digital processing of voxels
and 3D measurement functions and parametres.
Additional Physiology/ time series software, Topography software and Macro recorder/ editor software are also available.

  • Acousto Optical Tuneable Filter (AOFT) delivering exact control over laser lines and intensities with beam on and off with pin point accuracy
  • MMS-VIS Spectrometer, wavelenght range 320-950nm, Hamamatsu photodiode array detector

  • z series and 3D imaging, xyz scans
  • 4D imaging, xyzt scans
  • X-Z imaging, xz scans
  • reflected light imaging
  • transmitted light imaging

Images obtained from Confocal Microscope

Images show immunofluorescent staining of the cytoskeleton. The blue picture on the RHS shows microtubles. The image on the LHS is an overlay of this and actin (in red) and the Golgi apparatus (in green).
